A group of scientists from the Departamento de Fisiología, Facultad de Medicina, Universidad Nacional Autónoma de México, the Facultad de Estudios Superiores Zaragoza, Universidad Nacional Autónoma de México, and the Instituto Nacional de Geriatría, Secretaría de Salud, México City have published new insights into a protein called α-Klotho. This protein, long linked with longevity, appears to play a critical role in protecting the brain from the effects of aging, Alzheimer’s disease, Parkinson’s disease, and ischemic stroke.

According to this Medical News report, the study reveals that soluble Klotho not only has anti-aging properties but also acts as a strong anti-inflammatory and antioxidant shield for the brain. As levels of this protein naturally decline with age, the risk of neurodegeneration rises sharply, making it a promising target for future therapies.
 
How Klotho Works in the Brain
Klotho is mainly produced in the kidneys and the choroid plexus of the brain. Once released, the soluble form circulates in the blood and cerebrospinal fluid. It influences many processes including calcium balance, protection of brain cells from oxidative stress, and regulation of inflammatory pathways. Animal studies have shown that overexpression of Klotho extends lifespan and preserves memory, while deficiency accelerates aging-related decline.
 
The researchers describe how Klotho blocks harmful pathways such as NF-κB and the NLRP3 inflammasome, both of which drive inflammation in the brain. By reducing proinflammatory cytokines like IL-1β and TNF-α, Klotho helps preserve synaptic connections and prevent neuron death.
 
Relevance in Major Brain Diseases
In Alzheimer’s disease, Klotho reduces the build-up of toxic amyloid plaques and abnormal tau proteins. In Parkinson’s disease, higher levels of Klotho are linked to slower cognitive decline and reduced damage caused by α-synuclein protein aggregates. In stroke, animal experiments show that Klotho lessens brain injury by improving blood vessel function, decreasing oxidative stress, and limiting the size of infarcts.
 
Ways to Increase Klotho Levels
One encouraging part of the study is the evidence that Klotho levels can be boosted. Physical exercise, particularly aerobic activity, has been shown to raise Klotho in both animals and humans. Certain medications already approved for other conditions—such as statins, diabetes drugs like pioglitazone, and even vitamin D—also appear to stimulate Klotho production. Scientists are also exploring direct Klotho therapies and gene-based approaches, though these remain experimental.
