Laboratory tests showed that the extract significantly relaxed the arteries in rats when the inner layer of blood vessels was intact. This effect was largely due to enhanced nitric oxide signaling—a molecule known for helping blood vessels widen and improve blood flow. When the endothelium was removed or nitric oxide synthesis was blocked, the relaxation effect was drastically reduced, confirming that lotus seed extract acts through this natural vascular pathway.
 
Nitric Oxide and Calcium Channel Inhibition
Further experiments on human endothelial cells demonstrated that lotus seed extract boosted nitric oxide production without causing any cellular damage. This indicates that the extract is both potent and safe. The team also discovered that the extract prevents calcium from entering blood vessel walls, effectively blocking one of the main causes of vessel tightening and high blood pressure. When tested on rats, intravenous doses of the extract caused a measurable drop in systolic and diastolic blood pressure—showing a clear and dose-dependent hypotensive effect.
 
Computer Modeling Reveals Deep Biological Insights
To understand how the extract works at a molecular level, researchers from the University of Tsukuba in Japan conducted computer simulations showing that the active alkaloids from lotus seeds bind tightly to the endothelial nitric oxide synthase (eNOS) enzyme. This interaction, especially at the tetrahydrobiopterin (BH4) binding site, helps stabilize the enzyme and increase nitric oxide production, confirming a scientific basis for the observed blood pressure-lowering effects.
