Solove Research Institute (Columbus, USA), University of Pittsburgh Medical Center (USA), and Memorial Sloan Kettering Cancer Center (New York, USA) has revealed that the amount of arginine in the diet can dramatically influence how effectively the body's immune system fights both colorectal cancer and respiratory viruses, including SARS-CoV-2 and influenza. Arginine Controls a Key Immune Process
Arginine is an amino acid naturally found in foods such as meat, fish, dairy products, nuts, and legumes. The researchers discovered that when arginine levels become too low, the body loses important arginine-carrying transfer RNAs needed to build proteins. As a result, ribosomes stall while producing major histocompatibility complex class I (MHC class I) proteins, which are essential for displaying virus-infected or cancerous cells to killer T cells. Surprisingly, this effect depends on the specific genetic codons used within MHC class I genes, revealing a highly selective form of protein regulation.
 
Higher Arginine Improved Cancer and Viral Outcomes
Animal studies showed that restricting dietary arginine weakened immune responses, increased colorectal tumor formation, and reduced the body's ability to control influenza and SARS-CoV-2 infections. In contrast, increasing arginine intake through dietary supplementation, or genetically preventing arginine breakdown by deleting myeloid-specific arginase 1, significantly increased MHC class I protein production. This allowed immune cells to recognize and destroy abnormal cells more efficiently, reducing tumor growth and improving survival during respiratory viral infections. These protective benefits disappeared in mice lacking β2-microglobulin (B2m), confirming that MHC class I is central to the process.
 
Important Implications for Future Therapies
The findings suggest that arginine deficiency may silently weaken immune defenses in cancer patients and people battling viral infections. Rather than acting simply as a nutrient, arginine directly influences how immune-related proteins are manufactured, opening new possibilities for nutritional therapies that enhance immunity. While additional human studies are needed before dietary recommendations change, the research provides compelling evidence that carefully increasing arginine availability could become an important strategy for improving antiviral protection and reducing colorectal cancer risk by strengthening the body's own immune surveillance system.
